The quantitative PCR method is ideal for STI (Sexually Transmitted Infection) tests because it can detect even small amounts of pathogen DNA. When someone is infected with an STI-causing pathogen, even small quantities of the specific pathogen DNA can be detected in their body.
The DNA is isolated and purified from the sample. In qPCR, it is heated to separate the DNA double helix strands. Specific pathogen sequences (primers) bind to the resulting single strands of DNA. Through an enzyme-catalyzed reaction, building blocks are added to these primers to replicate the DNA. By repeating the process multiple times, large amounts of copies of the pathogen DNA are generated. The primers are specifically labeled and trigger certain fluorescence signals during qPCR. Based on these signals, it can be inferred whether pathogen DNA is present in the sample.
